VRAINS is a combination of the acronyms VR, AI, and NS, respectively stand for "Virtual Reality", "Artificial Intelligence" and "Network Systems". The series is set roughly 10 years in the future where the LINK VRAINS, a Virtual Reality Network has been integrated into the infrastrucuture of the world.

Yu-Gi-Oh VRAINS revolves around Yusaku Fujiki, a high-school aged hacker who doesn't like to stand out from the crowd. Yusaku Fujiki begins dueling in the LINK VRAINS system as "Unknown", later changing his name to "Playmaker". Yusaku's past is shrouded in tragedy after he fell victim to the insidious Hanoi Project. Yusaku is somewhat of a loner but has a close relationship to Cal Kolter (known as Shoichi Kusanagi in the Japanese version), who acts as Yusaku's shield for Playmaker and Souburner. Cal operates out of a food truck called Cafe Nom (Cafe Nagi in the Japanese version), where both Yusaku and Cal use as a base for hacking.

The Yu-Gi-Oh VRAINS anime series also showcases new gameplay elements from the Yu-Gi-Oh Trading Card Game. After the Link Strike Starter deck was introduced, a new way of play was implemented into the trading card game. Now, only one monster at a time can be summoned directly from a player's Extra Deck, which is then placed in of the two new zones in the middle of the playing field known as the "Extra Monster Zone".
